If the filesystem in question is ext2 (or ext3, for that matter), it
might be that instead of locking files using POSIX system calls, postfix
is changing the ext2/3 file attributes (the (u) attribute, undeletable),
which I doubt, because AFAIK, it would be extremely non-portable among
UNIXEN. However, give it a try:

chattr(1)

Of course, you can always have some buggy sectors on the media. Check
dmesg. Or try remounting the drive, if downtime is acceptable.
